Dietary Blueberry and Bifidobacteria Attenuate Nonalcoholic Fatty Liver Disease in Rats by Affecting SIRT1-Mediated Signaling Pathway

Abstract: NAFLD model rats were established and divided into NAFLD model (MG group), SIRT1 RNAi (SI group), blueberry juice (BJ group), blueberry juice + bifidobacteria (BJB group), blueberry juice + SIRT1 RNAi (BJSI group), and blueberry juice + bifidobacteria + SIRT1 RNAi groups (BJBSI group). A group with normal rats was a control group (CG). BJB group ameliorated NAFLD, which was better than BJ group (P < 0.05). The lipid accumulation was lower in CG, BJ, and BJB groups than that in MG, SI, BJSI, and BJBSI groups (P… Show more

“…Previously, we similarly demonstrated a significant relationship between BJ phytocompounds and molecular events related to apoptosis and expression of osteoclastogenic factors induced by oxidative stress and SIRT1 activation [33]. Indeed, dietary BBs increase SIRT1 levels in mammals [49,50], and SIRT1 overexpression is also related to the inhibition of osteoclastogenic factors [58,59]. Moreover, SIRT1 activity promotes osteogenic differentiation of mesenchymal stem cells and activation of RUNX-2 [46][47][48].…”

“…Indeed, SIRT1 is related to the regulation of osteogenic differentiation of tendon and mesenchymal stem cells [46,47], and it is a positive regulator of RUNX-2 [47,48]. Moreover, this enzyme is activated in mammals by dietary blueberry [49,50] and in osteocytes by BJ [33]. Initially, the BJ effect on SIRT1 levels in SaOS-2 cells in the presence or not of BSO-induced oxidative stress was studied.…”

“…This study investigated also the role of sirtuin type 1 (SIRT1), a class III histone deacetylase involved in the regulation of apoptosis [34] and activated in mammals by dietary blueberry [35,36]. Figure 5A shows that in our experimental conditions the starvation did not affect SIRT1 expression, which, on the contrary, was remarkably increased by BJ or BE treatment as compared to control and starved cells.…”

“…In general, genes involved in lipid synthesis ( srebp1 , srebp2 , acc , fasn , dgat2 and hmgcr ) and uptake ( ldlr , scrab1 and cd36 ) have up-regulated, while genes involved in lipid degradation ( pparα , cpt1 , hsl , atgl and hl ) and secretion ( abcg5/8 and mttp ) have down-regulated at mRNA or protein level (Ann et al, 2015; Gou et al, 2016; Hou et al, 2016; Kang et al, 2016; Komiya et al, 2016; Li et al, 2016; Liu et al, 2016; Ren et al, 2014; Shindo et al, 2010; Van Rooyen et al, 2011; Zhang et al, 2015a). Consistent with rodent models, the mRNA expression of genes involved in lipid degradation and secretion was decreased in tree shrews treated by approach 4 (Fig.…”
