Dietary Carotenoids in Head and Neck Cancer—Molecular and Clinical Implications

Abstract: Head and neck cancer (HNC) is one of the most common cancers in the world according to GLOBCAN. In 2018, it was reported that HNC accounts for approximately 3% of all human cancers (51,540 new cases) and is the cause of nearly 1.5% of all cancer deaths (10,030 deaths). Despite great advances in treatment, HNC is indicated as a leading cause of death worldwide. In addition to having a positive impact on general health, a diet rich in carotenoids can regulate stages in the course of carcinogenesis; indeed, stron… Show more

“…It has been hypothesized that carotenoids may exert indirect antioxidant activity by blocking IKK kinase activity and thereby allowing NF-kB sequestration in cytoplasm [ 191 ] ( Figure 2 ). Furthermore, in addition to the NF-kB pathway itself, carotenoids can also regulate the action of the MAPKs ERK, JNK, and p38, which have been identified to be involved in the promotion of NF-kB activation [ 192 , 193 ].…”

“…These changes may cause cancer cells to grow and spread in the body RB1 (The retinoblastoma gene) is a tumor suppressor gene that was first www.nature.com/scientificreports/ discovered in a rare ocular pediatric tumor called retinoblastoma (RB) 58 . The RB1 gene is essential for normal progression through the cell cycle and exerts part of its function through the family of transcription factors (E2F) and many other intermediaries 59 . In the absence of normal RB1, genomic instability and chromosomal aberrations accumulate, leading to tumor initiation, progression, and ultimately metastasis 60 .…”

“…Głównymi czynnikami etiologicznymi nowotworów krtani i gardła są używki (głównie tytoń), a ich występowanie w różnych populacjach wyjaśnia w dużej mierze zróżnicowanie zachorowalności obserwowane na całym świecie [Starska-Kowarska 2022]. Oprócz tych dwóch głównych czynników ryzyka dieta również wydaje się pełnić istotną rolę w określaniu ryzyka tych nowotworów.…”
