Diethylaminoethyl (DEAE) binding fraction from Taenia solium metacestode improves the neurocysticercosis serodiagnosis

Abstract: Neurocysticercosis (NC) is one of the most important diseases caused by parasites affecting the central nervous system. We fractionated by ion-exchange chromatography using diethylaminoethyl (DEAE)-sepharose resin the total saline extract (S) from Taenia solium metacestodes and evaluated obtained fractions (DEAE S1 and DEAE S2) by enzyme-linked immunosorbent assay (ELISA, n = 123) and immunoblotting (IB, n = 22) to detect human NC in serum. Diagnostic parameters were established by ROC and TG ROC curves for EL… Show more

“…Several studies have demonstrated the usefulness of the partial fractionation to obtain antigenic fractions with increased diagnostic parameters using other chromatographic approaches. Fractionation techniques [detergent Triton X-114 (Machado et al 2007; Gonçalves et al 2010), affinity for concanavalin A and jacalin (Nunes et al 2010; Oliveira et al 2010), ion-exchange chromatography with carboxymethyl sepharose and diethylaminoethyl sepharose (DEAE)] (Nunes et al 2013; Ribeiro et al 2014) lead to antigenic fractions with high potential in NCC immunodiagnosis using T. saginata or T. solium metacestodes.…”

“… 64 Diethylaminoethyl binding fraction from T. solium metacestode has been reported to improve the NCC serodiagnosis. 65 Recently, two magnetic immune-chromatographic tests for serologic detection of teniasis (based on rES33 antigen) and cysticercosis (based on the rT24H antigen) have shown high sensitivity and specificity in cases with ≥2 viable brain cysts. 66 A multiantigen print immunoassay is also being evaluated as a simple and effective tool to compare antibody responses to different cysticercosis and teniasis antigens and for rapid detection of T. solium cases.…”
