Diethylcarbamazine (DEC): immunopharmacological interactions of an anti-filarial drug

Abstract: Anti-parasitic drugs may achieve their therapeutic effect either by direct activity against the pathogenic organism, or by altering host factors which lead to parasite killing. In this review, we discuss the evidence for an indirect mode of action for one major anti-filarial drug, diethylcarbamazine (DEC). The interpretation most consistent with existing data is that DEC alters arachidonic acid metabolism in microfilariae and in host endothelial cells. These changes may result in vasoconstriction and amplified… Show more

“…[14][15][16] The effective microfilaricidal activity of AgNPs at relatively low concentration as reported in this study, with evidence of apoptotic induction in microfilariae, can project nanosilver as a potential drug adjuvant against lymphatic filariasis. Unlike DEC, which is effective against microfilariae only in vivo and not in vitro, 35 nanosilver promises greater therapeutic effectiveness. Remarkably, a recent report has demonstrated the therapeutic potential of AgNPs against leishmaniasis, another parasitic disease caused by Leishmania tropica.…”

“…Em alguns poucos casos, pode haver necessidade de se fazer uso de antitérmicos e analgégicos. No entanto, não há indicação para o uso de corticosteróides com a finalidade de minimizar ou de abolir tais reações 46 . As reações adversas localizadas estão associadas à morte dos vermes adultos e apresentam-se clinicamente como linfangite e adenite.…”

“…It is extremely effective against microfilariae of several parasites, capable of reducing the number of microfilariae in the circulation of an infected human by 60% within four minutes (32). It can also be used as a prophylactic against B. malayi in humans and has been shown to have activity against L3 larvae in cats (33,34). However, despite the remarkable efficacy of the drug in vivo, the means by which DEC eliminates B. malayi have not been established.…”
