Difference in viral binding between two epstein‐barr virus substrains to a spectrum of receptor‐positive target cells

Abstract: Radio-labelled Epstein-Barr virus (EBV) was utilized in a direct binding assay to detect the presence of EBV receptors. The sensitivity of this method was affirmed by the detection of EBV-receptors on three EV-carrying cell lines that have previously been reported as receptor negative. Two laboratory substrains of EBV, derived from the cell lines B95-8 and P3HR-I (designated B and P virus respectively), were tested in the binding assay. The main repcptor prototype adsorbed both viral strains without apparent d… Show more

“…HABPs 11402, 11403, and 11404 contain this reported epitope (underlined in the following: SAVTTPTPNATSPTLGKTSPT); HABPs 11401 and 11405 contain only part of this epitope (AVTTPTPNATSPT). The differences in cell binding tropism between B95-8 EBV (containing the repeat region) and P3HR-1 EBV (which does not contain this region) could be due to this binding region (46,47).…”

“…The transforming prototype B9.5-8 and P3HR-1 viruses were shown to bind to the same receptors on lymphoma cell lines [28]. When resting B cells were exposed to P3HR-1 virus first and thereafter to B95-8 virus the transformation was abrogated [32].…”

“…Virus binding and uptake assays were performed as described [28]. One million tonsillar B cells were incubated for 2 h at 37°C in the presence of 5000 cpm [3H]thymidine ([3H]dThd)labeled virus in a total volume of 1 ml medium with 10% fetal calf serum (FCS).…”
