Different culture media affect growth characteristics, surface marker distribution and chondrogenic differentiation of human bone marrow-derived mesenchymal stromal cells

Abstract: BackgroundBone marrow-derived mesenchymal stromal cells (BM-MSCs) play an important role in modern tissue engineering, while distinct variations of culture media compositions and supplements have been reported. Because MSCs are heterogeneous regarding their regenerative potential and their surface markers, these parameters were compared in four widely used culture media compositions.MethodsMSCs were isolated from bone marrow and expanded in four established cell culture media. MSC yield/1000 MNCs, passage time… Show more

“…First of all, equine MSCs from different sources were Original Article shown to display differences in growth kinetics (36,46), differentiation potential (36,47), and gene expression (36), thus differences in surface marker expression could be expected as well. Furthermore, differences in the culture medium could influence the cell characteristics (48,49). In this study, to enable obtaining sufficient cell numbers from all MSC sources, DMEM supplemented with 20% FBS was used, instead of 10% which is more commonly used (31,33,39).…”

“…It is known that culture medium may affect expression of cell markers [27][28][29]. We have used serum-free RPMI 1640 as a culture medium which should have minimal impact on amnion in vitro to mimic in vivo conditions.…”

“…Since tissue regeneration requires functional blood ves- In the absence of any universal and specific marker to define mesenchymal stem cells, their immunophenotyping relies on the use of combinations of both positive and negative markers. Note that MSCs profile may vary depending on the cell culture conditions [88] , or with their in situ localization [89] . Expression of the cell surface antigens CD73, CD90, CD105…”
