Different stages of B cell differentiation in non-T acute lymphoblastic leukemia.

Abstract: Abstract. Immunoglobulin heavy chain gene rearrangement was evaluated in 19 cases of acute lymphoblastic leukemia (ALL) and correlated with the immunological phenotypic expression on primary or phorbol diester (12-0-tetradecanoylphorbol-1 3-acetate[TPA])-induced cells. One case of common ALL (cALL), one case of T-ALL, and one undifferentiated acute leukemia that responded to anti-myeloid drugs after unsuccessful anti-lymphoid induction therapy, had germ line heavy chain genes. Rearranged immunoglobulin genes w… Show more

“…In contrast, the pattern for four of six B-cell lineage (LB) markers was the same in cALL as for B cells and/or B-cell ALL. These observations are consistent with and reinforce findings based on immunoglobulin gene rearrangement and cell-surface markers that suggest a B-cell origin of cALL (8,14,25).…”

“…Usually, 95% pure blasts were obtained. Restriction enzyme analysis, Southern blot electrophoresis and hybridization to a 32P-nick-translated probe were performed as previously described [31]. The probe used to detect TRGy rearrangements is a 0.7-kb Hind 111-Eco RI genomic fragment containing the J,1 region [27], which is located a short distance upstream of the C,1 gene [9].…”

“…The original stud- Receivedfor publication 4 March 1991 and in revisedform 22 October 1991. ies have been confirmed and extended by a large number of groups (2)(3)(4)(5). In most cases only one or two rearranged bands are seen (1); however, in some cases three or more rearranged bands can be identified (6)(7)(8)(9). Various explanations have been proposed; these range from the suggestion that all of the rearranged bands are present in all ofthe cells to the suggestion that the disease is in fact oligoclonal and that individual clones carry unique rearrangements.…”
