Differential actions of second messenger systems in the corpus luteum

Abstract: Background The corpus luteum develops from the follicular cells which remain following ovulation and secretes progesterone which is essential for normal pregnancy in domestic animals. In sheep, the corpus luteum must be present for at least 50 days if normal pregnancy is to be maintained (Casida & Warwick, 1945). Progesterone is produced in the corpus luteum of domestic animals by two morphologically and functionally distinct steroidogenic cell types termed large and small luteal cells. In sheep, large cells a… Show more

“…Luteinizing hormone stimulates adenylyl cyclase leading to increases in intracellular cAMP [7], activation of PKA, and ultimately stimulation of steroidogenesis [8, 9]. Both the small and large luteal cells respond to LH stimulation, however, the small luteal cells are many times more responsive to LH and activators of the cAMP/protein kinase A (PKA) signaling pathway [10, 11] when compared to large luteal cells.…”

“…Further, transcription of CAMKK2 was regulated by an increase in intracellular Ca 2+ [Okuno S et al, 1997]. Given that in the bovine CL, PGF 2α stimulates hydrolysis of phosphatidylinositol 4, 5-biphosphate and mobilizes intracellular Ca 2+ [Davis JS et al, 1987b;Wiltbank MC et al, 1991] these mechanisms of transcriptional regulation might be relevant. In support of the importance of CAMKK2 in the acquisition of luteal sensitivity to the luteolytic actions of PGF 2α is the finding that YWHAZ was down-regulated in D-10 CL.…”

“…YWHAZ has been shown to inhibit the activity of PKCs by interacting with a cystein-rich C1 domain [Robinson K et al, 1994;Toker A et al, 1990]. Importantly, PKCs mediate the luteolytic actions of PGF 2α [Wiltbank MC et al, 1991].…”
