Differential Effect of Urotensin II on Vascular Tone in Normal Subjects and Patients With Chronic Heart Failure

Abstract: In addition to direct effects on the myocardium, U-II may contribute to the increased peripheral vascular tone that is characteristic of human CHF. The present observations support the contention that the U-II system may be a potentially important target for pharmacological blockade in the treatment of this condition.

“…Interestingly, in healthy volunteers, a vasodilatory effect was observed, whereas a vasoconstrictive effect was observed in subjects with essential hypertension following UII administration. This same phenomenon was observed when the skin microvasculature tone was compared between healthy subjects and subjects with congestive heart failure: upon UII administration normal subjects displayed UII-mediated vasodilation, whereas patients with CHF displayed UII-mediated vasoconstriction (71). The reported variations in UII activity in diseased vs. healthy humans provides an important example of the difference between physiological and pathophysiological states and how the role of UII can drastically change in these distinct settings.…”

“…In a study by Lim et al (2004), a noninvasive iontophoresis technique was used to examine the Urotensin II and Its Receptor effects of UII on cutaneous blood flow in healthy and CHF patients. Although UII administration caused vasodilation in healthy patients, in patients with CHF, UII caused vasoconstriction, indicated by reduced blood flow in skin microcirculation.…”
