Differential effects of angiostatic steroids and dexamethasone on angiogenesis and cytokine levels in rat sponge implants

Abstract: 1 Subcutaneous implantation of sterile polyether sponges elicited a reproducible neovascular response in rats, as determined by blood flow measurement with a 133Xe clearance technique and confirmed histologically. This model was used to monitor the levels of two cytokines during angiogenesis and to compare the activities of angiostatic steroids and anti-inflammatory steroids. 2 Initial experiments followed the neovascular development over a 20-day period. Daily local injection of hydrocortisone caused a dose-d… Show more

“…24). High-dose prednisolone has been shown to suppress production of proinflammatory cytokines (25), to induce apoptosis (26), and to inhibit angiogenesis (27). Because of the progressive character of the pathophysiologic process that is the basis of radiologic damage, we believe that both the high initial dosage of prednisolone and its timing are crucial for achieving the effects seen in the COBRA group.…”

“…Values for PPE Pm and bromelain were not signifi cantly different from each other, but were signifi cantly lower than that for dexamethasone. In addition, some side effects, common to long-term glucocorticoid therapy, were evaluated: the immunosuppressive action, by weighing thymus and spleen (Ben Rhouma and Sakly, 1994;Hori et al, 1996), and the catabolic effect, by measuring the loss of body mass. While dexamethasone caused significant atrophy of spleen and thymus (50 and 86%, respectively), bromelain and PPE Pm only reduced thymus weight to the same low degree (26%).…”
