Differential Effects of Glycyrrhiza Species on Genotoxic Estrogen Metabolism: Licochalcone A Downregulates P450 1B1, whereas Isoliquiritigenin Stimulates It

Abstract: Estrogen chemical carcinogenesis involves 4-hydroxylation of estrone/estradiol (E1/E2) by P450 1B1, generating catechol and quinone genotoxic metabolites that cause DNA mutations and initiate/promote breast cancer. Inflammation enhances this effect by up-regulating P450 1B1. The present study tested the three authenticated medicinal species of licorice, [Glycyrrhiza glabra (GG), G. uralensis (GU), and G. inflata (GI)], used by women as dietary supplements, for their anti-inflammatory activities and their abili… Show more

“…8,32,4951 However, the AhR mediated P450 1A1/1B1 induction in MCF-10A cells is relatively low as reported previously 36 and confirmed in the present study. Data also suggest that the Phase I and Phase II enzyme expression is variable in MCF-10A cells depending on the confluence level in cell culture.…”

“…MCF-10A cells were cultured in 1:1 Dulbecco’s modified Eagle medium and Ham’s F12 nutrient mixture (DMEM/F12) with 15 mM HEPES and l -glutamine (Invitrogen), supplemented with 20 ng/mL epidermal growth factor, 100 ng/mL cholera toxin, 0.5 μg/mL hydrocortisone, 10 μg/mL insulin, 5% horse serum, and 1% penicillin–streptomycin at 37 °C with 5% CO 2 as described previously. 32 MCF-7 cells were maintained in RPMI 1640 media supplemented with 10% fetal bovine serum, 1% glutamax, 1% AB/AM, 1% nonessential amino acids, and 3 μg/mL insulin. All experiments were done with cells under 15 passages in phenol-red free media supplemented with charcoal stripped serum and the same other ingredients.…”

“…Licochalcone A, an anti-inflammatory [1][2][3][4], immunomodulatory [5,6], and antimicrobial [3,5,[7][8][9] polyphenol extracted from Glycyrrhiza inflata [1][2][3]10], triggers apoptosis or inhibits proliferation and migration of tumor cells [11], including those of cervical cancer [12], oral squamous carcinoma [7,9,[13][14][15], esophageal carcinoma [16], gastric cancer [4,17,18], hepatocellular carcinoma [2,19], malignant pleural mesothelioma [20], bladder determined photometrically at 405 nm. The absorption of the supernatant of erythrocytes lysed in distilled water was defined as 100% hemolysis.…”
