“…In a similar manner, we have shown that repeated administration of the prototypical 5-HT 1A receptor agonist 8-hydroxy-2-(di-npropylamino)-tetralin (8-OH-DPAT) reduced basal 5-HT levels in the rat hippocampus to a lesser extent than the first administration, indicating a rapid desensitization or internalization of the 5-HT 1A receptors in the raphe nuclei (Yoshitake et al, 2003c). In a following study (Yoshitake and Kehr, 2004), it was demonstrated that the (R)-and (S)-optical isomers and the (R,S)-racemate of 8-OH-DPAT differentially reduced 5-HT release in the ventral hippocampus of awake rats. Thus, (R)-8-OH-DPAT (0.3 mg/kg s.c.) and the (R,S)-racemate caused a marked reduction of 5-HT levels, to 41 and 46%, respectively, whereas (S)-8-OH-DPAT (0.3 mg/kg s.c.) caused maximal reduction of 5-HT levels only to 74% of controls in both perfusion groups, as shown in Fig.…”
