Differential Leukocyte Count Method for Bovine Low Somatic Cell Count Milk

Abstract: Whereas many differential leukocyte count methods for high somatic cell count (SCC) milk from mastitic cows are available, only a few have been developed for low SCC milk. We have developed a flow cytometric differential leukocyte count method for low SCC milk. The procedure consists of 1) 1.5 ml of diluted milk sample (30%, vol/vol dilution with PBS), 2) centrifugation, 3) leukocyte labeling with SYTO 13 and 4) flow cytometric analysis. Four major leukocyte populations can be clearly identified in the green f… Show more

“…Epithelial cells were reported to be the major cell type, on the basis of absence of labeling with anti-CD11a/CD18 antibodies, in the milk of healthy cows [97,98]. Others found a majority of lymphocytes in milk by flow cytometry, particularly in early lactation [47]. The differential leukocyte count in low SCC milk samples is difficult, and the methods used differ widely.…”

“…(ii) Staining with commercial SYTO 13 green fluorescent nucleic acid stain (Molecular Probes, Eugene, Oregon, USA) was used as described by Dosogne et al (2003) in a slight modification: 490 µl of cell suspension in RPMI 1640 was stained with 10 µl of diluted (1:40) SYTO 13 solution.…”

“…Apoptotic and secondary necrotic apoptotic neutrophils were enumerated by FCM (FACScan, Becton Dickinson Immunocytometry Systems, San Marcos, Ca, USA). Two different staining procedures were used: Simultaneous staining with Annexin V labelled with fluorescein isothiocyanate (FITC) and propidium iodide (PI) as described by Vermes et al (1995) and SYTO 13 staining as described by Dosogne et al (2003).…”

“…The commercial SYTO 13 green fluorescent nucleic acid stain (Molecular Probes, Eugene, OR, USA) was used as described by Dosogne et al (2003) in a slight modification: 490 µl of cell suspension in RPMI 1640 was stained with 10 µl of diluted (1:40) SYTO 13 solution. After incubation at laboratory temperature in the dark for 15 min, the specimens were placed on ice and stored up to the FCM measurement.…”
