Differential Reovirus-Specific and Herpesvirus-Specific Activator Protein 1 Activation of Secretogranin II Leads to Altered Virus Secretion

Abstract: Viruses utilize host cell machinery for propagation and manage to evade cellular host defense mechanisms in the process. Much remains unknown regarding how the host responds to viral infection. We recently performed global proteomic screens of mammalian reovirus TIL-and T3D-infected and herpesvirus (herpes simplex virus 1 [HSV-1])-infected HEK293 cells. The nonenveloped RNA reoviruses caused an upregulation, whereas the enveloped DNA HSV-1 caused a downregulation, of cellular secretogranin II (SCG2). SCG2, a m… Show more

“…3.5 | Immunohistochemistry and effects of IL-6 on the expression of SCG2 protein and mRNA (Berard, Severini, & Coombs, 2015;Courel et al, 2014). To verify the results of screening DEGs in transcriptome sequencing, we first assessed SCG2 immunoreactivity in cancer and paracancerous tissues.…”

“…SGIV injected in muscle of grouper needs time to reach and infect cells of spleen and liver, while direct infection of SGIV in FHM cell activates JNK1 early after infection (more quickly). Phosphorylation and activation of JNKs has crucial impacts on the viral infection (Nacken et al, 2014 ; Berard et al, 2015 ). The localization of Ec-JNK1 showed no changes during SGIV infection.…”

“…The dual phosphorylation motif TPY of JNK is important for transferring signals from the cell membrane to nuclear transcription factors, by increasing their ability to activate transcription factors (Kyriakis et al, 1994 ; Dong et al, 2002 ; Stahl et al, 2012 ). Virus seems to activate the JNK-AP-1 pathway to promote infection (Kumar et al, 1998 ; Holloway and Coulson, 2006 ; Berard et al, 2015 ). The JNK signal pathway represents a major physiological mechanism of regulation of c-Jun that relates to AP1 (Gupta et al, 1996 ; Davis, 2000 ).…”

“…Secretogranin II (SCG2), a key AP‐1‐regulated member of the granin family, is a neuroendocrine protein that regulates the biogenesis of secretory granules involved in hormonal peptide sorting [ 16 ], mediates neuronal differentiation, and protects neuroblastoma cells from nitric oxide‐induced apoptosis. SCG2 is cooperatively activated by glutamate‐dopamine hippocampal neurons as a potential regulator of food intake and non‐chronic bodyweight change by interacting with secretogranin III and accumulating appetite‐related hormones into secretory granules [ 17 ].…”
