“…In contrast to the in vitro mature-embryo culture of other cereals, as of barley (He and Jia 2008) and rice (Li et al 2015), the rate of somatic embryogenesis of the analyzed wheat species, with the exception of T. diccocum, was low, and they were unable to form more than one green plant per initial explant ( Table 2). The callus regeneration ability of all cultures of AB genome tetraploid wheats, except for T. turgidum, was higher (11.0-28.6%) than that of hexaploid (2.3-8.8%) and diploid wheats (0.7-6.1%).…”

“…For complementation tests, the CDS of BOS1 fused with a green fluorescent protein (GFP) and the 1,795-bp promoter upstream of ATG were cloned into the pCambia 1300 binary vector with Xba I/ Bam HI and Pst I/ Spe I enzymes, respectively. Then, the BOS1p : BOS1c - GFP plasmid was transformed into a bos1-1 mutant through the agrobacterium ( Agrobacterium tumefaciens )-mediated transformation of rice mature embryos ( Li et al., 2015 ; Zhu et al., 2021 ). In the second and third filial transgenic generations (T 2 and T 3 ), the phenotypes of the complementary transgenic lines harboring BOS1p : BOS1c - GFP construction were observed.…”

“…For the complementary test, a 1464 bp MIL1/OsHPL3 CDS fused to Green Fluorescence Protein (GFP) driven by the Cauliflower Mosaic Virus (CaMV) 35S constitutive promoter was cloned into the pCambia1300 binary vector. Then, the resulting 35S:MIL1c-GFP plasmid was introduced into the mil1-1 mutant via the agrobacterium ( Agrobacterium tumefaciens )-mediated transformation of mature rice embryos as described previously [ 39 , 43 ]. In the T 2 and T 3 generations, we observed the phenotypes of complementary transgenic lines containing the 35S:MIL1c/OsHPL3c-GFP construct.…”
