Differentially Methylated Epiloci Generated from Numerous Genotypes of Contrasting Tolerances Are Associated with Osmotic-Tolerance in Rice Seedlings

Abstract: DNA methylation plays an essential role in plant responses to environmental stress. Since drought develops into a rising problem in rice cultivation, investigations on genome-wide DNA methylation in responses to drought stress and in-depth explorations of its association with drought-tolerance are required. For this study, 68 rice accessions were used for an evaluation of their osmotic-tolerance related to 20% PEG6000 simulated physiological traits. The tolerant group revealed significantly higher levels of to… Show more

“…This is in accordance with our finding that dose-insensitive salt-induced DMMs appeared more commonly in sites that could facilitate perturbation in the expression of genes that hypothetically could be part of a molecular response to salinity, as reported elsewhere [65]. There is evidence from previous studies to suggest that salt-induced DMMs can play important roles in evoking metabolic differences between seedlings growing under control and saline conditions [25,26,29,34,64,66], although the markers found in these works were either few in number or else identified from a single genotype. The provision here of a robust list of consistent, salt-responsive DMMs therefore provides a useful starting point from which to gather a more holistic picture of DNA methylation-mediated regulation of molecular responses to salt exposure.…”

“…All transgenic plants and WPB106 (wild type, WT) were grown in the greenhouse at 28–35 °C, in Shanghai, or in fields at the station of the Shanghai Academy of Agricultural Sciences under normal growth conditions. For salinity stress at the seedling stage, seedlings of rice were cultivated in normal nutrient solution for 5 days after germination on a 96-well plate (Xia et al 2017 ). They were placed in a growth chamber (14 h of daytime at 30 °C and 10 hat night at 20 °C with 70% relative humidity).…”

“…Seedlings of WA-CMS and its maintainer line were transplanted to a 96-well plate 3 days after germination and cultivated in a nutrient solution (Xia et al, 2017 ). They were placed in a growth chamber (14 h of light at 30°C and 10 h of darkness at 20°C with 70% relative humidity).…”
