Differentiation of cardiac Purkinje fibers requires precise spatiotemporal regulation of Nkx2‐5 expression

Abstract: Nkx2-5 gene mutations cause cardiac abnormalities, including deficits of function in the atrioventricular conduction system (AVCS). In the chick, Nkx2-5 is elevated in Purkinje fiber AVCS cells relative to working cardiomyocytes. Here, we show that Nkx2-5 expression rises to a peak as Purkinje fibers progressively differentiate. To disrupt this pattern, we overexpressed Nkx2-5 from embryonic day 10, as Purkinje fibers are recruited within developing chick hearts. Overexpression of Nkx2-5 caused inhibition of s… Show more

“…It has been found that Nkx-2.5 plays critical roles in regulating tissue-specific gene expression and induces chicken cardiac myocyte differentiation in tissues (Schultheiss et al, 1995;Harris et al, 2006). Cyclic adenosine monophosphate response element-binding protein, which binds to the cyclic adenosine monophosphate response elements and increases or decreases the transcription of genes, is critical in a variety of cellular processes such as cell proliferation, differentiation, and adaptive responses in chickens (Sirotkin and Grossmann, 2003;Kim et al, 2005).…”

“…In addition, investigators have also demonstrated a role for Nkx.2.5 in specification of the peripheral conduction system in a time- and dose-dependent manner 26, 27 . Nkx2.5 haploinsufficiency causes hypocellularity of the peripheral conduction system.…”

“…In addition, there was a severely limited range of interrogation angles for Doppler imaging due to the egg shell. These limitations were largely overcome by using an ex ovo culture setup (Dunn et al, 1981;Tuan, 1983;Ono and Tuan, 1986;Harris et al, 2006), which allowed much freer positioning of the scanhead (Fig. 1) and additionally re- Using an ex ovo culture setup, unhindered access is obtained, and multiple scanning interrogation angles are possible.…”
