Differentiation of Xanthomonas albilineans strains with monoclonal antibody reaction patterns and DNA fingerprints

Abstract: Thirty-eight strains of the sugarcane leaf scald pathogen, Xanthomonas albilineans, were compared using a series of seven species-specific monoclonal antibodies as well as genomic DNA fingerprint patterns. The strains, which were obtained from 13 countries and states in the USA, were placed in three major groups with approximately eight subgroups based on similarity of their serological reactions. The serological groupings correlated strongly with groupings based on DNA fingerprint patterns. The groups of stra… Show more

“…A large genetic diversity exists among worldwide strains of X. albilineans, including three serovars and six lysovars of the pathogen (Rott et al, 1986(Rott et al, , 1994. Serological diversity of X. albilineans has been confirmed using a combination of monoclonal antibodies and DNA fingerprinting (Alvarez et al, 1996). Using pulsed-field gel electrophoresis (PFGE) and the rare-cutting enzyme SpeI, more than 200 strains of the pathogen were distributed in 10 genetic groups (PFGE-A to PFGE-J; Davis et al, 1997;Champoiseau et al, 2006).…”

“…In addition, the method using pronase (Alvarez et al, 1996;Hartung & Civerolo, 1987) takes from 4 to 6 hours, the method described by Pan et al (1997) is simpler but requires water bath and takes about three hours to DNA isolation the DNA. The last described protocol (Ausubel et al, 1992; (1992).…”

“…Several procedures are described for Xanthomonas DNA isolation (Alvarez et al, 1996;Hartung & Civerolo, 1987), as well as for Pseudomonas spp. and Erwinia spp., where the bacteria are grown in Wilbrink's agar plates for 72h and, cells are treated with egg white lysozyme for 30 min on ice.…”

“…Considerando a diversidade de perfis de banda, diferentes variantes foram identificados entre isolados originários da Flórida, em áreas onde se verificou a ocorrência de quebra de resistência de cultivares de cana (2). Também com base nos perfis de bandas de DNA, foi demonstrada a presença de variação genômica intra-específica em isolados coletados em diversos países e em distintas regiões de um mesmo país (8).…”
