Direct Cytotoxicity of Lentinula edodes Mycelia Extract on Human Hepatocellular Carcinoma Cell Line

Abstract: Lentinula edodes mycelia (L.E.M.) is a dried powder extracted from shiitake mushrooms (Lentinula edodes). We previously demonstrated that it has immunomodulatory effects. In this paper, the direct cytotoxic effects of the polysaccharide-rich fraction of L.E.M. (L.E.M. ethanol precipitate; LEP) on HepG2 human hepatocellular carcinoma (HCC) cells were investigated. LEP directly killed the HepG2 cells efficaciously, but had only minor effects on normal rat hepatocytes and normal mouse dermal cells under the same … Show more

“…Present studies revealed that BE3 consists mainly of ribonucleic acid (59.1%). 59,60 Sine the anticancer action of FA-2-b-β and LEM are in the same vein as changes caused by the BE3 fraction, it may also confirm the importance of ribonucleic acids presented in BE3 for its chemoprevention abilities. 54,55 Among the mentioned studies, the detailed molecular mechanism of the anticancer action was described only by Sun et al 54 They discovered that FA-2-b-β (RNAprotein complex isolated from A. blazei) in combination with AZT (3′-azido-3′-deoxythymidine) induced apoptosis in the human gastric cancer cell line MKN45 which was associated with inhibition of tumor cell telomerase as well as BCL2 down-regulation and up-regulation of caspase 3 mRNA.…”

“…The low temperature (<50°C) aqueous extract of this mushroom significantly decreased proliferation of the human tumor cell lines laryngeal carcinoma (HEp-2) and cervical adenocarcinoma (HeLA), with IC 50% values ranging from 0.46% to 1.03% [13]. Yukawa et al, [22] examined the antiproliferative effect of L. edodes mycelia extract on HepG2 human hepatocellular carcinoma cells. The extract efficaciously induced apoptosis of tested type of cells through the caspase -3 and -8 death receptor pathways [22].…”

“…Mushrooms are a good source of natural compounds for cancer treatments. Many edible mushrooms and their products have been noted as dietary supplements and sources of medicinal compounds with potential anticancer properties against several types of cancer in human (Ferrari et al, 2012;Yukawa et al, 2012. ) L. edodes is one of the medicinal mushrooms which was studied and showed numerous pharmacological properties, such as antimicrobial, antiviral, antioxidant, anti-inflammatory, antiatherogenic, hypoglycemic, hepatoprotective, anticancer, and immunomodulating properties (Ferreira et al, 2012;Rai et al, 2005;Zaidman et al, 2005).…”

Several compounds extracted from L. edodes such as Lentinan (Chihara et al, 1970;Zhang et al, 2007), the polysaccharide L-II (Zheng et al, 2005), water extracts of fruiting bodies (Israilides et al, 2008), L•E•M (Shen et al, 2009), and LEP (Yukawa et al, 2012), show immunomodulatory and anticancer activity.…”
