Direct enantioselective HPLC monitoring of lipase‐catalyzed kinetic resolution of flurbiprofen

Abstract: The solvent versatility of Chiralpak IB, a 3,5-dimethylphenylcarbamate derivative of cellulose-based chiral stationary phase, is demonstrated in the direct enantioselective HPLC monitoring of lipase-catalyzed kinetic resolution of flurbiprofen in nonstandard HPLC organic solvents. Nonstandard HPLC organic solvents were used as the reaction media for the lipase-catalysis and in mean time as diluent to dissolve the "difficult to dissolve" enzyme substrate (the acid) and as eluent for the simultaneous enantiosele… Show more

“…Furthermore, it should be considered, that lipases tolerate a great number of non-natural substrates, are stable and active in organic solvents and necessitate no cofactors. Based on the previous reports, it seems to be obvious, that use of these enzymes as biocatalysts in the industry will be continuously increasing due to their high activity and low price [6][7][8][9].…”

“…Candida antarctica lipase is commercially available as a free and immobilized form. Additionally, what is important from a pharmaceutical point of view, CALB shows a stereopreference towards the ( R )‐enantiomer in organic solvents which was reported and proven in many studies . However, in some cases the enantiopreference of CALB is towards the ( S )‐enantiomer, which is dependent on the support on which the enzyme is immobilized as well as the reaction medium .…”
