The actual organization of the central nucleus of the amygdala (CEA) in the rat is mostly based on cytoarchitecture and the distribution of several cell types, as described by McDonald in 1982. Four divisions were identified by this author. However, since this original work, one of these divisions, the intermediate part, has not been consistently recognized based on Nissl-stained material. In the present study, we observed that a compact condensation of retrogradely labeled cells is found in the CEA after fluorogold injection in the anterior region of the tuberal lateral hypothalamic area (LHA) in the rat. We then searched for neurochemical markers of this cell condensation and found that it is quite specifically labeled for calbindin (Cb), but also contains calretinin (Cr), tyrosine hydroxylase (TH) and methionine-enkephalin (Met-Enk) immunohistochemical signals. These neurochemical features are specific to this cell group which, therefore, is distinct from the other parts of the CEA. We then performed cholera toxin injections in the mouse LHA to identify this cell group in this species. We found that neurons exist in the medial and rostral CEAl that project into the LHA but they have a less tight organization than in the rat.

A previous study indicated that the swallowing reflex is inhibited during rhythmic jaw movements induced by electrical stimulation of the anterior cortical masticatory area. Rhythmic jaw movements were induced by electrical stimulation of the central amygdaloid nucleus (CeA). The swallowing central pattern generator is the nucleus of the solitary tract (NTS) and the lateral reticular formation in the medulla. Morphological studies have reported that the CeA projects to the NTS and the lateral reticular formation. It is therefore likely that the CeA is related to the control of the swallowing reflex. The purpose of this study was to determine if rhythmic jaw movements driven by CeA had inhibitory roles in the swallowing reflex induced by electrical stimulation of the superior laryngeal nerve (SLN). Rats were anesthetised with urethane. The SLN was solely stimulated for 10 s, and the swallowing reflex was recorded (SLN stimulation before SLN + CeA stimulation). Next, the SLN and the CeA were electrically stimulated at the same time for 10 s, and the swallowing reflex was recorded during rhythmic jaw movements (SLN + CeA stimulation). Finally, the SLN was solely stimulated (SLN stimulation following SLN + CeA stimulation). The number of swallows was reduced during rhythmic jaw movements. The onset latency of the first swallow was significantly longer in the SLN + CeA stimulation than in the SLN stimulation before SLN + CeA stimulation and SLN stimulation following SLN + CeA stimulation. These results support the idea that the coordination of swallowing reflex with rhythmic jaw movements could be regulated by the CeA.

Further, Shirasu et al [47] have shown that anterograde-labeled terminal buttons on Vmes neuronal somata are more abundantly present in the caudal than in the rostral Vmes after a wheat germ agglutinin conjugated to horseradish peroxidase injection into the CeA. The same study also indicated that a portion of these terminal buttons form axo-somatic synapses with Vmes neurons, and that the CeA has direct projections to the Me5, suggesting that the amygdala regulates bite strength by modifying neuronal activity in the Vmes.…”

The organization of emotional motor behavior including jaw movements is governed by neural circuits of the limbic system, such as the amygdala and hypothalamus. GABAergic neurons in the central amygdaloid nucleus (CeA) exert an inhibitory influence on premotor neurons for the trigeminal motor nucleus (Vm) in the parvicellular reticular formation (RFp) of the medulla oblongata. The CeA also influences glutamatergic posterior lateral hypothalamic neurons and non-dopaminergic neurons in the retrorubral field of the midbrain, both of which send their axons to Vm-premotor neurons in the RFp. In addition, the CeA may modulate the activity of Vm motoneurons via projections to the mesencephalic trigeminal nucleus whose neurons convey inputs from the masticatory muscle spindles and periodontal ligament receptors to jaw-closing motoneurons in the Vm. These pathways from the amygdala to the trigeminal motor system in the lower brainstem may underlie the regulation of emotional jaw movement. Figure 1. Various forms of sensory inputs that become emotional stimuli are sent to the amygdala via the thalamus and association cortices as well as via the brainstem nuclei. The context of these emotional stimuli is sent to the amygdala via the hippocampal formation. The amygdala integrates this information and evaluates the biological value of emotional stimuli. Subsequently, subjective experience of emotion and expression of emotion are induced. The expression of emotion consists of emotional behaviors, autonomic responses, and endocrine responses.
