Direct Regulation of Gonadotrophin‐Releasing Hormone (GnRH) Transcription By RF‐Amide‐Related Peptide‐3 and Kisspeptin in A Novel GnRH‐Secreting Cell Line, mHypoA‐GnRH/GFP

Abstract: RF-amide-related peptide-3 [RFRP-3; also often referred to as the mammalian orthologue of the avian gonadotrophin-inhibitory hormone (GnIH)] and kisspeptin have emerged as potent modulators of neuroendocrine function via direct regulation of the reproductive axis in the hypothalamus and pituitary. There are few studies focusing on the direct regulatory effects of RFRP-3 and kisspeptin on gonadotrophin-releasing hormones (GnRH) neurones. We report their effect on GnRH mRNA expression and release in a novel GnRH… Show more

“…This finding suggests that GnIH might be involved in central inhibition by restraining pulsatile secretion of GnRH during the prepubertal period. This hypothesis is supported by evidence that GPR147 is expressed on GnRH neurons [ 36 , 37 ]. Furthermore, administration of GnIH or its mammalian ortholog, RFRP3, has been reported to result in inhibition of GnRH secretion [ 9 , 38 40 ].…”

“…GnIH (RFRP3) also suppresses the firing of kisspeptin‐activated vGluT2 (vesicular glutamate transporter 2)‐GnRH neurons as well as of kisspeptin‐insensitive GnRH neurons . Suppression of GnRH mRNA expression by GnIH (RFRP3) administration was also shown in a novel GnRH neuronal cell model, mHypoA‐GnRH/GFP . Accordingly, GnIH may inhibit the secretion of gonadotropins by decreasing the activity of GnRH neurons in addition to directly regulating pituitary gonadotropin secretion.…”

“…A study using another GnRH neuronal cell model, mHypoA-GnRH/GFP, generated from adult-derived GnRH-GFP neurons, shows the interaction of GnIH and kisspeptin on GnRH transcriptional regulation (65). In mHypoA-GnRH/GFP cells, treatment of GnIH attenuates basal GnRH mRNA expression, whereas kisspeptin induces GnRH levels.…”

“…Co-treatment of GnIH and kisspeptin suppresses GnRH mRNA expression, suggesting the inhibitory effect of GnIH may override the stimulatory effect of kisspeptin on GnRH mRNA expression. By using transcriptional inhibitors (actinomycin D and DRB), Gojska et al further show that GnIH-mediated repression is involved in new RNA synthesis rather than affecting the stability of pre-existing GnRH mRNA in mHypoA-GnRH/GFP cells (65). Although they present a novel action mechanism of GnIH on GnRH transcriptional regulation, these results do not indicate the direct inhibitory effect of GnIH on kisspeptin-induced signaling pathway.…”
