The general rules established from mammalian species for the regulation of spermatogenesis by gonadotropins may not be fully relevant in fish. Particularly, Fsh is as potent as Lh to stimulate steroidogenesis and the Fsh receptor is expressed in Leydig cells. In seasonal breeders, Fsh is likely the major gonadotropin involved in spermatogenesis onset and Lh is required to support spermatogenesis progression and gamete release. However, the genes that relay the action of Fsh and Lh have been poorly investigated in fish. The present study was aimed at identifying gonadotropin-dependent genes expressed in the testis during fish puberty. We cultured pubertal trout testicular explants for 96 h, with or without gonadotropin, and analyzed transcriptome variations using microarrays. Fsh and Lh had similar effects on a large group of genes while other genes were preferentially regulated by one or the other gonadotropin. We showed that most of the responsive genes were expressed in somatic cells and exhibited relevant patterns during the seasonal reproductive cycle. Some genes preferentially modulated by Lh could be involved in testicular cell fate (pvrl1 and bty) or sperm maturation (ehmt2 and racgap1) and will deserve further examination. Besides Fsh's effects on the steroidogenic pathway, our study demonstrates that Fsh coordinates relevant stimulatory and inhibitory paracrine factors known to regulate early germ cell proliferation and differentiation. Some of these genes belong to major regulatory pathways including the Igf pathway (igf1b/igf3 and igfbp6), the Tgfb pathway (amh, inha, inhba, and fstl3), the Wnt pathway (wisp1), and pleiotrophin (mdka).

It has been more than 20 years since the first batch of transgenic fish was produced. Five stable germ-line transmitted growth hormone (GH) transgenic fish lines have been generated. This paper reviews the mechanisms of integration and gene targeting of the transgene, as well as the viability, reproduction and transgenic approaches for the reproductive containment of GH-transgenic fish. Further, we propose that it should be necessary to do the following studies, in particularly, of the breeding of transgenic fish: to assess the fitness of transgenic fish in an aqueous environment with a large space and a complex structure; and to develop a controllable on-off strategy of reproduction in transgenic fish. transgene, integration mechanism, GH transgenic fish, population fitness Citation:Hu [6,7]. In cooperation with Hunan Normal University, we produced a transgenic triploid fish with a growth rate that is 15% faster than that of the control carp, with a forage utilization rate increased by 11.1%. The transgenic triploid fish is completely sterile [8]. Such completely sterile transgenic triploid carp such as the all-fish GH transgenic carp may be suitable for industrialized conditions. The US Food and Drug Administration (FDA) is evaluating the ecological effects of current and potential uses of the field release of GH transgenic Atlantic salmon, which would promote the commercial farming of transgenic fish. No transgenic animals have been released into a natural environment for commercialized cultivation as food. Transgenic fish would be the breakthrough model as the first commercial farming transgenic animal. We review the mechanisms of integration and controllable regulatory expression of transgenes and the population fitness assessments of GH transgenic fish. We will conclude by providing suggestions for the thorough study of genetic engineering in breeding fish for potential commercial uses.

Muscovy duck (Cairina moschata) is characterized by broodiness, and egg-laying traits in this species should be of primary concern. As a member of the IGF superfamily of proteins, insulin-like growth factor-1 (IGF-1) is an important positive regulator of growth and gonad development in vertebrates. To explore the effect of IGF-1 on the growth hormone/insulin-like growth factors (GH/IGF) axis, we cloned and characterized IGF-1 of Muscovy duck. The expression level of IGF-1 was widely expressed in all the tested tissues, and the highest level was detected in the liver. In laying ducks, IGF-1 expression levels in the hypothalamus, pituitary, and ovary were very significantly higher (P < 0.01) than that in the tissues of nesting duck, and IGF-1 expression levels in the heart and liver were significantly higher (P < 0.05) than that in nesting duck tissues. Furthermore, a single nucleotide polymorphism (SNP) (A/G) was found and significantly (P < 0.05) associated with age at first egg and egg number at 300 d. This study provides the first evidence that IGF-1 promote egg-laying traits of Muscovy duck through two axes, involving GH/IGF and hypothalamic-pituitary-gonadal axes. These findings enrich the information of IGF-1 of Muscovy duck and demonstrate for the first time the ability of IGF-1 to promote reproduction, indicating that IGF-1 could be used as an important marker gene.
