Discovery of a novel putative atypical porcine pestivirus in pigs in the USA

Abstract: Pestiviruses are some of the most significant pathogens affecting ruminants and swine. Here, we assembled a 11 276 bp contig encoding a predicted 3635 aa polyprotein from porcine serum with 68 % pairwise identity to that of a recently partially characterized Rhinolophus affinis pestivirus (RaPV) and approximately 25-28 % pairwise identity to those of other pestiviruses. The virus was provisionally named atypical porcine pestivirus (APPV). Metagenomic sequencing of 182 serum samples identified four additional A… Show more

“…VIDISCA-454, a next generation sequencing technique with unbiased viral RNA amplification, was performed on the serum. The sequence reads showed some identity to pestiviruses at the time of first analysis (2012), though a significant similarity to APPV was apparent once the genome of this pestivirus became available in GenBank (March 2015; [16]; Figure 1A). Via genome walking, the full length sequence of the viral genome was obtained (APPV NL1).…”

“…This novel pestivirus has been first described by Hause et al [16]: atypical porcine pestivirus (APPV). Arruda et al [15] inoculated the virus in pregnant sows and the fetal amniotic vesicles of these sows.…”

“…Porcine sapelovirus and porcine kobuvirus were each detected in a single pool. The recently described atypical porcine pestivirus was also identified in a single pool (Hause et al, 2015). A final virus showing~90 % sequence similarity to murine pneumonia virus (MPV; formerly pneumonia virus of mice) and canine pneumovirus (CPV) was identified in two pools.…”
