Discovery of microRNA-like Small RNAs in Pathogenic Plant Fungus Verticillium nonalfalfae Using High-Throughput Sequencing and qPCR and RLM-RACE Validation

Abstract: Verticillium nonalfalfae (V. nonalfalfae) is one of the most problematic hop (Humulus lupulus L.) pathogens, as the highly virulent fungal pathotypes cause severe annual yield losses due to infections of entire hop fields. In recent years, the RNA interference (RNAi) mechanism has become one of the main areas of focus in plant—fungal pathogen interaction studies and has been implicated as one of the major contributors to fungal pathogenicity. MicroRNA-like RNAs (milRNAs) have been identified in several importa… Show more

“…These results are challenging to obtain and frequently require follow-up studies to confirm functionality and role of a proposed mi/milRNA [58]. Evidence of cleavage found in degradome and 5'-RACE experiments hints that fungal mi/milRNAs may function more similarly to plants [31,[59][60][61][62][63][64][65], as this evidence indicates directed cleavage of a target RNA. However, much more information is needed to develop a detailed view of these dynamics, especially in terms of base-pairing requirements [66] or distinct mechanisms of different AGOs [11].…”

“…In fungi, mature miRNAs are produced from miRNA precursors by Dicer enzymes 30 ; thus, fungal miRNAs should be referred to as microRNA-like RNAs (milRNAs). MilRNAs play important roles in regulating fungal gene expression and pathogenicity mechanisms under different environmental conditions 30 – 32 . The predicted targets of lncRNAs demonstrate their potential regulatory roles and the study of lncRNAs related to aflatoxin biosynthesis and fungal metabolism may enhance our understanding of the regulatory functions of lncRNAs under different stress conditions.…”
