Discovery of Potent Small-Molecule Inhibitors of Ubiquitin-Conjugating Enzyme UbcH5c from α-Santonin Derivatives

Abstract: As a therapeutic target for antitumor necrosis factor (TNF)-α interventions, UbcH5c is one of the key ubiquitin-conjugating enzymes catalyzing ubiquitination during TNF-α-triggered nuclear factor kappa B (NF-κB) activation. In the present study, three series of analogues were designed and synthesized from α-santonin, and their UbcH5c inhibitory activities were screened by Western blotting and NF-κB luciferase assay. Further BIAcore, in-gel fluorescence imaging, and immunoprecipitation assays demonstrated that … Show more

“…The surface of the sensor chip was regenerated via the injection of 10 μL of the regeneration buffer (5 mM NaOH). The association rate constant k a and dissociation rate constant k d were calculated and analyzed using the monovalent analyte model, and the equilibrium dissociation constant (KD) was calculated (KD = k d /k a ) [29].…”

“…To date, approximately twenty inhibitors of ubiquitin-conjugating E2 enzymes have been discovered (Ardecky et al, 2010; Ceccarelli et al, 2011; Chen et al, 2017; Cheng et al, 2014; Cornwell et al, 2019; Garg et al, 2020; Helms et al, 2011; Kothayer et al, 2016; Liu et al, 2014; Pulvino et al, 2012; Sanders et al, 2013; Strickson et al, 2013; Wang et al, 2018, 2011). Many of the reported inhibitors target the active site Cys, others act by disrupting protein-protein interactions, and some have an allosteric effect on ubiquitin transfer.…”

“…Diverse butyrolactones have been studied to evaluate anti-inflammatory activities (Entry 1-9 in Table 2). Some of these butyrolactones modulate the NF-κB signaling pathway such as a santonine-derived butyrolactone that showed anti-inflammatory activity through the inhibition of the ubiquitin-conjugating enzyme, UbcH5c (Entry 1 in Table 2) [23,24]. This anti-inflammatory activity was maintained in vivo using Freund's adjuvant arthritis rat model.…”
