Discovery of Sulfated Small Molecule Inhibitors of Matrix Metalloproteinase-8

Abstract: Elevated matrix metalloproteinase-8 (MMP-8) activity contributes to the etiology of many diseases, including atherosclerosis, pulmonary fibrosis, and sepsis. Yet, very few small molecule inhibitors of MMP-8 have been identified. We reasoned that the synthetic non-sugar mimetics of glycosaminoglycans may inhibit MMP-8 because natural glycosaminoglycans are known to modulate the functions of various MMPs. The screening a library of 58 synthetic, sulfated mimetics consisting of a dozen scaffolds led to the identi… Show more

“…For this, we evaluated the electrostatic surface potential (ESP) of the three studied proteins, as shown in Fig. 3 66 . The presence of multiple electropositive micro-domains being close to each other could favor the binding of small molecule inhibitors.…”

“…Plasmatic MMP8 concentrations were linked to the presence and severity of coronary artery disease [ 193 ] and serum MMP8 levels were associated with cardiovascular outcome in patients [ 194 ]. The recent discovery of small molecule inhibitors of MMP8 such as sulfated mimetics of glycosaminoglycans (sulfated quinazolinones) looks promising to prevent atherosclerosis-mediated cardiovascular events [ 195 ].…”
