Discrimination among cultivars of rapeseed (Brassica napus L.) using DNA polymorphisms amplified from arbitrary primers

Abstract: RAPDs (Randomly Amplified Polymorphic DNAs) were used to discriminate among 23 cultivars of oilseed rape (Brassica napus) selected from several breeding programs. A set of 100 random sequence 10-mer primers were tested, of which 70 produced bands and 22 showed evidence of polymorphism. A selection of six primers produced 23 polymorphic bands of between 300 to 2200 base pairs in size, sufficient to distinguish between the cultivars. An analysis of seed of five cultivars obtained from four different sites showed… Show more

“…However, phenotype is influenced by the environment and its interaction with a particular genotype. Molecular DNA techniques such as random amplified polymorphic DNA (RAPD) analysis have been applied to detect genetic variation in various crops (Mailer et al 1994;Demeke et al 1996). RAPD analysis has the advantage of identifying polymorphisms independent of both prior DNA sequence knowledge and environmental influence.…”

“…PCR was performed using a PTC-100™ (MJ Research, Inc.), as reported by Mailer et al (1994). RAPD products were separated by electrophoresis on ethidium bromide stained 1.4% agarose gels in 1× TAE buffer.…”

“…Molecular markers have successfully been applied in registration activities of cultivars (Mailer et al, 1994) and also identification of varieties as a part of seed and grain trade (Bligh et al, 1999). A powerful technique for DNA fingerprinting is the Polymerase Chain Reaction (PCR) amplification of tandem repeat sequences.…”

“…Hence, the proposed plant variety protection lends added urgency to the search for solutions to the conservation of plant genetic diversity. Molecular markers have been successfully applied in registration activities including cultivar identification (Mailer et al, 1994). However, with the necessity of varietal identity, present study was an attempt to reveal genetic diversity and varietal identification of eight cotton varieties in Bangladesh by DNA fingerprinting using microsatellite markers.…”
