Disruption studies of a Candida albicans gene, ELF1: a member of the ATP-binding cassette family

Abstract: A 3.6 kb gene (ELF7) with homology to the ATP-binding cassette (ABC) gene family has been isolated from genomic libraries of Candida albicans. Members of this gene family include both membrane transport proteins which confer a drug-resistance phenotype, and proteins whose functions are associated with protein translation. ELF7 (Elongation Like Factor) showed greatest homology with a Sacchammyces cemwisiae ORF (YPu26w), whose function is unknown, and lower homology with fungal elongation factor 3 (EF-3) genes. … Show more

“…In some instances, a rapid protocol was used to isolate genomic DNA (Burke et al, 2000). Southern analysis was performed as previously described (Sturtevant et al, 1998). Blots were probed with the C. albicans BMH1 ORF isolated from plasmid pJP1.…”

“…These clones were manually sequenced and one clone that showed remarkable homology to the 14-3-3 proteins (Muhlschlegel et al, 1998;Sturtevant, 2000) was used to screen an EcoRI C. albicans SC5314 Lambda Zap II library (Sturtevant et al, 1998). A 5 kb EcoRI genomic fragment in pBluescriptSK + phagemid was excised from a tertiary positive plaque from the Lambda Zap II SC5314 library.…”

“…Genomic DNA from C. albicans was isolated using glass beads (9). Southern blot analysis was performed as previously described (43). Probes were radiolabeled on purified plasmid inserts with [␣-32 P]dCTP by using Ready-To-Go DNA Labeling Beads (Amersham Pharmacia Biotech).…”
