Dissimilar Characteristics of N-Methyl-N-Nitrosourea-Initiated Foci and Tumors Promoted by Dichloroacetic Acid or Trichloroacetic Acid in the Liver of Female B6C3F1 Mice

Abstract: Dichloroacetic acid (DCA) and trichloroacetic acid (TCA) are metabolites of the industrial solvent and environmental contaminant trichloroethylene (TCE), as well as contaminants of chlorinated drinking water. Human exposure to these chemicals is of concern as all three have been shown to increase liver tumor incidence in mice. Differences in dose-response curves, progression to cancer, and postexposure regression of lesions suggest that TCA and DCA work through different mechanisms. The purpose of this study w… Show more

“…In passing, it should be noted that the consistency of c-Jun expression noted in DCA-promoted liver tumors in MNUinitiated mice (143) was much lower than previously reported in tumors induced in male mice (117). This could be a sex difference, but it is important to recognize that different antibodies were used in these two studies.…”

“…A CDAA diet triggered severe fibrosis and enzyme‐altered preneoplastic foci at 12 weeks through the generation of oxidative stress in rat liver due to strong N2‐guanine tRNA methyltransferase II activity and relative resistance to DNA hypomethylation . In contrast, no discernible preneoplastic lesion was found at 22 weeks after CDAA diet feeding because of the lack of a sensitive marker for small tumor foci in mice, as typified by the glutathione S‐transferase placental form in rats . Preneoplastic foci of altered hepatocytes were observed at 65 weeks in mice fed CDAA diet .…”

“…Furthermore, the phenotypes of the foci and tumors induced by DCA and TCA are different (Latendresse and Pereira, 1997;Stauber and Bull, 1997;Tao et al, 1998Tao et al, , 1996. Tumors induced by DCA are c-Jun positive and glycogen poor, whereas TCA-induced lesions do not display these characteristics (Stauber and Bull, 1997), suggesting a different mechanism of action.…”
