Dissolution Test for Silymarin Tablets and Capsules

Abstract: Silybine (SBN), isosilybine (ISBN), silycristine (SCN), silydianine (SDN), and taxifoline (TXF) are the main active flavonoids commonly found in the dried fruits of Silybum marianum, Gaertner (Compositae). Concentrations of these compounds, except TXF, are usually expressed together as silymarin content. This paper describes a simple dissolution test developed to estimate silymarin (Sl) in pharmaceutical formulations. Five commercial products were tested using this new method (including tablets, sugar tablets,… Show more

“…Modifications included a methanol : water:acetic acid (35 : 60 : 5) mobile phase and UV detection at 277 nm. This preparation of silymarin was comparable to several commercial formulations characterized by Campodónico et al [11]. Mice were given five daily intraperitoneal (i.p.)…”

“…Silymarin was purchased from Sigma-Aldrich Chemical Company (St. Louis, MO). This silymarin consists of a mixture of seven isomers including taxifolin (4 %), silichristin (27.9 %), silidianin (2.9 %), silybin A (19.3 %), silybin B (31.3 %), isosilybin A (8.2 %) and isosilybin B (2.3 %) as determined by high performance liquid chromatography as described previously [11]. Modifications included a methanol : water:acetic acid (35 : 60 : 5) mobile phase and UV detection at 277 nm.…”

“…The stationary phase, Bondclone C 18 column (300 mm x 3.9 mm, 10 µm -Phenomenex, CA, USA), was kept at 25°C. The mobile phase was a mixture of methanol : double distilled water : acetic acid (50:50:1) 21 . The flow rate was 1.0 ml/min and the run time was 30 min.…”

“…Silybin is one of the major flavonolignans from the mixture & consists of about 60-70% of silymarin. Silybin is a mixture of two diastereomers (A, B) which are present in a ratio of 1:1 [1,2]. Silybin is a known hepatoprotectant but due to its low serum, tissue levels, low solubility & absorption lead to its poor oral bioavailability [3,4].…”
