Distinct signaling by insulin and IGF-1 receptors and their extra- and intracellular domains

Abstract: Insulin and insulin-like growth factor 1 (IGF-1) receptors share many downstream signaling pathways but have unique biological effects. To define the molecular signals contributing to these distinct activities, we performed global phosphoproteomics on cells expressing either insulin receptor (IR), IGF-1 receptor (IGF1R), or chimeric IR-IGF1R receptors. We show that IR preferentially stimulates phosphorylations associated with mammalian target of rapamycin complex 1 (mTORC1) and Akt pathways, whereas IGF1R pref… Show more

“…Hence, one possible explanation for our data is a ligand-independent role of tip cell insulin receptors. Indeed, recent data from Ronald Kahn’s group indicate a ligand-independent role of insulin receptors in the membrane trafficking of brown pre-adipocytes ( 46 ), potentially aligning with our findings. However, although the concentrations of insulin experienced by sprouting endothelial cells in vivo are unknown, our in vitro data are likely to reflect low picomolar concentrations of insulin because of its presence in fetal calf serum.…”

“…Insulin receptor substrates (IRSs), which are the adaptor proteins of the IR, recruit multiple signaling complexes [ 111–114 ]. In particular, growth factor receptor-bound protein 2 (Grb2) is recruited to the binding motif on IRS [ 115 , 116 ], which in turn forms a complex with guanine nucleotide exchange factor Son of Sevenless and phosphorylates RAS. Activated RAS (RAS-GTP) activates the mitogen-activated protein kinase (MAPK) signaling cascade, including extracellular signal-regulated kinase 1/2 (ERK1/2) [ 117 , 118 ].…”

“…Insulin can also bind to the insulin-like growth factor 1 (IGF1) receptor [ 116 , 128 , 129 ], which consequently activates the mitogenic signaling pathways that promotes cellular growth and proliferation. Although there is significant redundancy in the intracellular insulin and IGF signaling pathways, some studies imply that they may have distinct roles in malignancies.…”

“…Decades of research have defined distinct actions of insulin and IGF-I in vivo , but attempts over many years to relate this to differences in signalling architecture have instead established that the signalling networks activated by the receptors are closely similar. Application of modern tools has suggested there are some quantitative differences in activation of particular elements of the network ( Cai et al, 2017 ; Nagao et al, 2021 ; Liu et al, 2012 ), although many studies have used unphysiologically high ligand concentrations, sometimes orders of magnitude above interstitial concentrations. Collective findings, to date, suggest that ligand availability, tissue receptor expression profiles, expression of intracellular signalling components and, perhaps, distinct signalling dynamics may underpin the different biological roles of the receptors.…”
