Distinct transcriptional regulatory mechanisms underlie left-right asymmetric expression of lefty-1 and lefty-2

Abstract: Both lefty-1 and lefty-2 genes are expressed on the left side of developing mouse embryos and are implicated in left-right (L-R) axis formation. With the use of transgenic analysis, the transcriptional regulatory regions of these genes responsible for their L-R asymmetric expression have now been investigated. The 9.5-kb upstream region of lefty-1 and the 5.5-kb upstream region of lefty-2 reproduced the expression pattern of the corresponding gene. Examination of deletion constructs revealed the presence of a … Show more

“…In the Cerl2(sGFP-D3 0 -UTR) BAC, the 3 0 -UTR of Cerl2 was replaced by the polyadenylation signal sequence (250 bp) of SV40. All transgenic mice were established as previously described 37 . In some cases (when indicated as F0 assay), transgenic embryos were directly examined before establishing stable transgenic lines.…”

“…Lefty locus contains two genes with the same transcriptional orientation in human, mice, and zebrafish. Human Lefty1 is identical to mice LeftyB and Lefty2 is identical to human LeftyA (10)(11)(12)(13). LeftyB has 96% sequence identity with LeftyA and these two proteins differ only in 16 amino acids.…”

“…Lefty2 is expressed in the left LPM and is a direct target of Nodal (Meno et al, 1997;Meno et al, 1996;Saijoh et al, 1999;Saijoh et al, 2000). Lefty1 is expressed in the left side of the PFP but, unlike Nodal, Lefty2 and Pitx2c, its expression is independent of Foxh1.…”
