“…Additionally, DA depletion caused no significant alterations in the striatal GDNF protein levels of MPTPtreated rhesus monkeys (Collier et al, 2005). In contrast, there are several studies that do detect an upregulation of the striatal GDNF mRNA and protein levels after administration of 6-OHDA (Zhou et al, 2000;Nakajima et al, 2001;Yurek and Fletcher-Turner, 2001;Katoh-Semba et al, 2007). However, there are some important differences in experimental setup compared to our study.…”

“…Animals were sacrificed 18 days post-lesioning since increases in GDNF protein level have been described at 2-3 weeks after administration of 6-OHDA in different animal models of Parkinson's disease (Nakajima et al, 2001;Yurek and Fletcher-Turner, 2001;Katoh-Semba et al, 2007).…”

“…In concurrence with previous reports saying that GDNF is mainly produced by and secreted from astrocytes during embryonic development [33,34], we found that GDNF is mostly expressed in GFAP-positive cells after AAV2-GDNF gene transfer in the striatum, suggesting that astrocytes are still a main source for GDNF synthesis, although some studies showed that the rAAV-EGFP-transduced cells in the striatum were mainly neurons in appearance [35,36]. In our study, these GDNF-labeled astrocytes remained in the SVZ and DG after AAV2-GDNF gene transfer at least for 4 weeks, and PH3+NeuN- and BrdU+NeuN-positive cells in these two regions were further enhanced as compared with the control mice injected with AAV2-EGFP, implying that GDNF transgene can induce neuroregeneration in the SVZ and DG.…”

“…In a previous study with double-labeling of the substantia nigra using TH immunohistochemistry, associated with GDNF in situ hybridization, furthermore, Pochon et al 1997 show that the majority of nigral neurons express GDNF. A more recent study also demonstrates that GDNF is normally synthesized in neurons, but may be caused by glial cells in damaged brains (Katoh-Semba et al 2007). Therefore, it is conceivable that BDNF and GDNF may play a key role in the maturation of neurons, interneurons and glial cells in the substantia nigra during postnatal development in mice.…”
