Distribution of Menaquinones in Actinomycetes and Corynebacteria

Abstract: SUMMARYMenaquinones were the only isoprenoid quinones found in 48 corynebacteria and actinomycete strains examined. Dihydromenaquinones having nine isoprene units were the main components isolated from Gordona, Mycobacterium, Corynebacterium bovis, Corynebacterium glutamicum and a strain labelled Nocardia farcinica, but dihydromenaquinones having eight isoprene units were characteristic of other Corynebacterium species and representatives of the ' rhodochrous' complex. Tetrahydromenaquinones having six and eig… Show more

“…The type of diaminopimelic acid (DAP) isomers was determined by the method of Becker et al [11]. Menaquinones were extracted and purified by the methods of Collins et al [12], then analyzed by HPLC equipped with a CAPCELL PAK C18 column (Shiseido) [13].…”

“…Biomass for the molecular systematics and chemotaxonomic studies was obtained after cultivation in yeast extract-glucose broth (yeast extract 1.0%, glucose 1.0%, pH 7.0) for 1 week at 27 1C. After extraction of isoprenoid quinones, as described by Collins et al, 26 analysis was done by HPLC (Agilent 1100, Agilent, Santa Clara, CA, USA) and mass spectrometer (JEOL JMS-T 100LP) using a Pegasil ODS column (Senshu, Tokyo, Japan), according to Tamaoka et al 27 Isomers of diaminopimelic acid were determined by TLC using whole-cell hydrolysates. 28 The N-acyl types of muramic acid were determined by the method of Uchida and Aida.…”

“…Diaminopimelic acid [11] and the composition of cell-wall sugars [12] was identified by TLC of the whole-organism hydrolysate. Fatty acids were determined by GC-MS [13,14], and menaquinones by reversed-phase HPLC [15,16]. 16S rRNA gene amplification and determination of the almost complete sequence (1495 bp) were carried out according to Rainey et al (1996) [17].…”
