Distribution volume, metabolic clearance and plasma half disappearance time of exogenous luteinizing hormone releasing hormone in normal women and women with obesity and anorexia nervosa

Abstract: Synthetic LRH was infused into normal women and women with obesity and anorexia nervosa to determine the distribution volume (DV), metabolic clearance rate (MCR) and half disappearance time (t\m=1/2\)of plasma LRH.In normal women, the DV of LRH was 12.1 \m=+-\0.9 (mean \ m=+-\se) 1, the MCR was 1478.9 \m=+-\39.8 ml/min (28.5 \m=+-\1.2 ml/min/kg body weight) and the initial t \ m=1/ 2\ was 5.6 \m=+-\0.4 min.In obese patients the DV (20.6 \ m=+-\ 1.5 1) was significantly higher than that in normal subjects (P < … Show more

“…The decline of MCR across puberty suggests that it is the rate of hypothalamic GnRH secretion rather than ambient sex steroid levels that determines MCR (15) which is also consistent with the lack of influence of androgen (44) or estrogen (18) on MCR in males, although the effects of estrogen treatment on GnRH MCR in hypogonadal women remain inconclusive (47). These effects of age and physiology on GnRH MCR may influence choice of appropriate GnRH and analog doses.…”

“…The first phase is a rapid (ti/ 2 2-8 min) composite of mixing throughout the vascular and extracellular fluid spaces with rapid clearance. When the mixing phase is eliminated in postinfusion studies of plasma disappearance (13)(14)(15)(16)(17), the initial half-lives are slightly longer (5-8 min). The second phase is a slower clearance component (half-times of 15-60 min) the physical interpretation of which remains unclear.…”
