Disulfiram, an old drug with new potential therapeutic uses for human cancers and fungal infections

Abstract: Disulfiram, a drug used to treat alcoholism, has recently been indicated to play a primary as well as an adjuvant role in the treatment of many cancers and in the reversal of fungal drug-resistance. This review discusses the molecular mechanism of action of disulfiram and its potential use in the treatment of human cancers and fungal infections.

“…In addition to acting as an aldehyde dehydrogenase inhibitor, DSF has been shown to inhibit, e.g., DNA topoisomerases, matrix metalloproteinases, and ABC drug transport proteins, and thereby to have antitumor and chemosensitizing activities (23,24). Monensin is a Na + /H + antiporter with antibiotic and antimalarial activities.…”

“…In this study, we report that tumor suppressive effects of DSF on leukemic cells depend on cell density, and we describe mechanism of this phenomenon. We show that DSF can significantly limit viability of human myeloid leukemia U937 and ML-1 cells by inducing caspase-dependent programmed cell death pathways in doses 100-200 ng/ml that are safe in human plasma (26). This effect is clearly cell density-dependent because it is most prominent in cells seeded at low density (50,000 cells/ml) and disappears as cell density increases.…”

“…Disulfiram, a member of dithiocarbamate family, has been widely used as an antialcoholism drug in the clinics for .60 years. 3 Recently, it has been demonstrated to possess strong antitumor ability for various tumors, including NSCLC. encapsulation of disulfiram with high drug loading, and meanwhile, they were not suitable for treating lung cancers owing to their low delivery ability to lungs in an oral or intravenous administration.…”
