Disulfiram enhanced delivery of orally administered copper into the central nervous system in Menkes disease mouse model

Abstract: Our data demonstrated that disulfiram enhanced the delivery of orally administered copper into the central nervous system in MD model mice. The administration of disulfiram will enable patients to avoid unpleasant subcutaneous copper injection in the future.

“…Kalor et al utilized parenteral copper ion injection (subcutaneous copper chloride-dithiramine) for Menkes disease treatment, but the injected copper ions were sequestered by the blood-brain barrier and did not exert a neurodegenerative effect ( Kaler, 2014 ). Hoshina et al conducted an intriguing study employing ^64Cu-labeled copper to visualize copper absorption and investigate the influence of DSF as a chelating agent on Cu absorption( Hoshina et al, 2018 ). In this study, DSF was administered prior to copper ion intake.…”

“…Mice with confirmed infections were treated with a single drug. Each drug was administered at the highest non-toxic concentration that has previously been used in mice (Sakamoto et al, 1984;Ulrich et al, 1989;Hoshina et al, 2018;Nakayama et al, 2018) (Table 1). The drug concentration in the serum was calculated using the modified Kirby Bauer assay (MKB).…”
