Context: Verbascum nigrum L. (Scrophulariaceae) is a perennial plant used in folk medicine for the treatment of kidney diseases due to its presumable diuretic properties. Objective: We investigated the diuretic activity and toxicity of extracts from different parts of V. nigrum and identified a group of compounds responsible for the biological effect. Materials and methods: Five ethanol extracts from herb, roots, flowers, leaves and stems as well as five fractions of polar compounds isolated from herb of V. nigrum were orally administrated as a single dose of 50 mg/kg to rats. Urinary excretion and electrolyte content were measured at 3 and 6 h after the treatment. The acute toxicity of the V. nigrum extracts and fractions was evaluated in mice. Results: All extracts, except the one prepared from the roots, showed a significant increase of the urine output within first 3 h after their administration. The extract from stems was the most active, inducing urine output of 14.6 AE 0.8 ml/kg BW versus 5.2 AE 1.4 ml/kg BW of the control. It also demonstrated saluretic activity with a natriuretic index 4.1 and a kaliuretic index 3.8. The diuretic activity was correlated with the flavonoid content in the plant organs. Flavonoid fractions demonstrated significant activity; the higher content of flavonoids (expressed as hesperidin) translated into more pronounced diuretic (35.9 AE 2.1 ml/kg BW) and saluretic effects (natriuretic index 4.5 and kaliuretic index 5.4). Conclusion: The diuretic activity of traditionally used V. nigrum was validated experimentally. The pharmacological effect was attributed to flavonoids, which accumulated in aerial parts of the plant, mainly in stems.

Callus induction and indirect organogenesis protocol were developed using different explants of Orthosiphon stamineus such as leaf, petiole, and internode on MS medium supplemented with different plant growth regulators. For callus induction, MS medium with 2, 4-D, NAA and IBA alone or in combinations with BAP were used. High-efficiency callus induction was obtained (100%) in petiole compared with other explants at the concentrations of 5mg/l 2, 4-D hormone, and 99% callus induction was obtained in leaf and internode at the concentration of 4.0 mg/l 2,4-D hormone. Similarly, 100% callus induction was obtained at the concentrations of 4.0 mg/l NAA with 0.5 mg/l BAP combinations in petiole and internode whereas 99% of callus induction was obtained from the same concentration. The induced callus of leaf, petiole, and internode was transferred to shooting medium fortified with plant growth regulators at 5.0 mg/l BAP and 0.5 mg/l NAA. Among these, internode has shown maximum percentage of the shoot (60%) regeneration than petiole and leaf callus. Shoot regenerated plantlets were then transferred to ¼MS medium fortified with 1.0 mg/l NAA hormone results in high efficient rooting (20) in vitro plantlets without inducing basal callus. Further, root-induced healthy plantlets were subjected to hardening in the greenhouse and established in natural environmental condition with a survival rate of 95% without any morphological changes of true type plants. Hence the developed protocol was optimized to yield a high frequency of callus induction and regeneration in different explants of O.stamineus with different concentrations of plant growth regulator.

Diuretic Action of AqueousOrthosiphonExtract in Rats

Abstract: An aqueous extract of Orthosiphonis folium, given orally, enhances considerably ion excretion in rat to a level comparable to that obtained with furosemide. No aqueretic action is observed. The increased ion excretion is not due to the potassium content of the starting material.
