Diverged Binding Specificity of Rim101p, the Candida albicans Ortholog of PacC

Abstract: The biology of Candida albicans, including dimorphism and virulence, is significantly influenced by environmental pH. The response to ambient pH includes the pH-conditional expression of several genes, which is directly or indirectly regulated by Rim101p. Rim101p is homologous to PacC, a transcription factor that regulates pH-conditional gene expression in Aspergillus nidulans. PacC binds 5-GCCARG-3 sequences upstream of pH-responsive genes and either activates or represses transcription. The absence of pacC c… Show more

“…Our results could indicate that PacC regulation of Bcacp1 and Scacp1 proceeds through another DNA motif. One such motif exists in Candida albicans (59-CCAAGAA-39; Ramon & Fonzi, 2003), but it is absent in the Bcacp1 and Ssacp1 promoters. Alternatively, the results could indicate that Bcacp1 and Ssacp1 are not regulated via PacC, as shown for several genes in C. albicans (Davis et al, 2000).…”

“…In order to construct the conditional mutant, the BMH1 ORF with appropriate 39 flanking sequence was cloned into pCIp10-mal2 (provided by Dr M. Stark; Cassettes containing mutated BMH1 alleles. The plasmid pLUX was used as a template vector for construction of BMH1 plasmids (Ramon & Fonzi, 2003). pLUX contains the URA3 gene, and when digested with NheI allows complete reconstitution of the URA3 locus in strain BWP17; it was kindly provided by Dr W. A. Fonzi (Georgetown University).…”
