Diversity and Co-Occurrence Patterns of Fungal and Bacterial Communities from Alkaline Sediments and Water of Julong High-Altitude Hot Springs at Tianchi Volcano, Northeast China

Abstract: The Julong high-altitude volcanic hot springs in northeast China are of undeniable interest for microbiological studies due to their unique, extreme environmental conditions. The objective of this study was to provide a comprehensive analysis of the unexplored fungal and bacterial community composition, structure and networks in sediments and water from the Julong hot springs using a combination of culture-based methods and metabarcoding. A total of 65 fungal and 21 bacterial strains were isolated. Fungal gene… Show more

“…all had best hits that originated in Arctic sediments ( F. petrolei , Chaudhary et al, 2019 ) or thawing permafrost samples ( F. psychrolimnae , Aszalós et al, 2020 and F. degerlachei , Zeglin et al, 2016 ) ( Table 3 and Supplementary Table 3 ). Pseudomonas extremaustralis (sq55) was also detected in Arnarnesstrýtan vent fluids and has previously been observed in other alkaline hot springs ( Table 3 ; Wang and Pecoraro, 2021 ). Furthermore, this ASV (Sq55_Pseduomonas) was not detected in background seawater samples.…”

“…Microscopy was done using a Nikon ECLIPSE Ci-L microscope (Tokyo, Japan) to examine fungal morphological characters, such as hyphae, pseudohyphae, conidiophores, conidia, poroconidia, arthroconidia, etc. ( Wang and Pecoraro, 2021 ). All isolated fungal strains were deposited in the LP Culture Collection (personal culture collection held in the laboratory of Prof. Lorenzo Pecoraro), at the School of Pharmaceutical Science and Technology, Tianjin University, Tianjin, China.…”

“…Fungal DNA sequences were submitted to GenBank under accessions ON711628-ON712642. Fungal morphology was characterized based on macroscopic and microscopic observations ( Pecoraro et al, 2020 ; Wang and Pecoraro, 2021 ). Microscopic morphological traits, such as septate hyphae, phialides, hyphal coils, conidiophores, conidia, and conidial chains were examined under a Nikon ECLIPSE Ci-L microscope (Tokyo, Japan) for identification of isolates following the taxonomic keys for different taxa ( Gilman, 1957 ; Lucas, 1981 ; De Hoog et al, 2000 ).…”
