DNA Barcode for Identifying Folium Artemisiae Argyi from Counterfeits

Abstract: Folium Artemisiae Argyi is an important herb in traditional Chinese medicine. It is commonly used in moxibustion, medicine, etc. However, identifying Artemisia argyi is difficult because this herb exhibits similar morphological characteristics to closely related species and counterfeits. To verify the applicability of DNA barcoding, ITS2 and psbA-trnH were used to identify A. argyi from 15 closely related species and counterfeits. Results indicated that total DNA was easily extracted from all the samples and t… Show more

“…DNA barcoding has been demonstrated to be an efficient tool for identifying traditional medicines as well as their original species [23,[58][59][60]. In the present study, this tool was used to discriminate among the five original species of Fritillariae cirrhosae bulbus and three of its adulterants.…”

“…However, these loci have not been analyzed in the wide range of taxa of the Asteraceae family [40]. Additionally, known core chloroplast barcodes, such as rbcL, matK, rpoB and rpoC1, had low discriminatory power for classification of Artemisia taxa [21][22][23]. Therefore, discovery of novel barcodes with high discriminatory power or use Universal barcode primer set designed for angiosperm [30] of the whole plastome as a super-barcode is important.…”

“…Recently, the subgenus Pacifica, including Hawaiian species, was recognized by nuclear ribosomal (ITS and ETS) and chloroplast (trnL-F and psbA-trnH) markers [20]. However, the resolution of these markers was insufficient to resolve taxonomic issues at the species level due to high sequence similarity of closely related taxa presumably caused by rapid radiation and hybridization [21][22][23][24]. Therefore, development of novel DNA markers or barcodes for investigation of Artemisia is an important challenge.…”

“…They are widely used as dietary functional material, food flavoring and coloring agent in China, Japan, Korea and other East Asian countries. [1][2][3] They can also be used both orally and externally for medical purposes, such as channeled to the liver, spleen and kidney. 4) They are proved to have various clinical effects, e.g., meridian warming, bleeding stopping, cold dispersing, pain relieving, and miscarriage preventing.…”
