DNA-based genealogy reconstruction of Nebbiolo, Barbera and other ancient grapevine cultivars from northwestern Italy

Abstract: Northwestern Italy is a wine region of the world with the highest of reputations, where top quality wines of remarkable economic value are produced from traditional, long-cultivated varieties. Kinship analyses were performed using 32 microsatellite loci and more than 10 K single-nucleotide polymorphism markers on 227 traditional grapes mostly from Northwestern Italy—including those that have been neglected or are threatened. This was done to better understand the genetic grapevine origins and history of this r… Show more

“…The mentioned founders, the historic “Fortana” and the SD related “Spergola” (alias “Vernaccia di Oristano”) are all quoted as early as XVII century on the Apennine foothills ( D’Onofrio et al, 2015 ; Bignami et al, 2015a ). This would support the Northern Apennines area as the likely homeland for all these grapes, including “Spergola”, later introduced to Sardinia as already asserted on the base of other genetic considerations ( Raimondi et al, 2020 ).…”

“…The software COLONY enables pedigree reconstruction using a likelihood-based method. The main limitation of this approach is related to its high computation demand that restricts its application to relatively small datasets (see, e.g., Crespan et al, 2020 ; Raimondi et al, 2020 ). The pedigree reconstruction method adopted in our study had the advantage of being applicable to a very large dataset, since it is not based on computation intensive likelihood estimations.…”

“…V. vinifera ‘Nebbiolo’ is one of the most important and ancient grape vines in the world [ 23 , 24 ]. Its area of cultivation is predominantly limited to northwestern Italy (hilly and mountainous areas of Piedmont, Aosta Valley, and Lombardy), although today it is also cultivated in California, Mexico, Australia, and other countries.…”

“…Significant efforts have been made to recover and identify local minor grapevine germplasm in many Italian regions, like Apulia, Campania, Emilia Romagna, Friuli Venezia Giulia, Liguria, Sardinia, Sicily, Tuscany (Costantini et al, 2005;De Mattia et al, 2007;Torello Marinoni et al, 2009;Carimi et al, 2010;Crespan et al, 2011;De Lorenzis et al, 2014;Schneider et al, 2014;Mercati et al, 2016;D'Onofrio et al, 2016;Gristina et al, 2017;Pastore et al, 2020), and variety pedigrees have been reconstructed for the most widespread and renowned ones and for those having local importance, helping to unravel the intricate skein of the structure of Italian grapevine populations (Di Vecchi-Staraz et al, 2007;Ruffa et al, 2016;Crespan et al, 2020;Raimondi et al, 2020;D'Onofrio et al, 2021). Similar efforts have been made in the Marche region, because the resulting knowledge is very useful in science and has cultural and marketing appeal.…”
