DNA demethylation affects imprinted gene expression in maize endosperm

Abstract: Background DNA demethylation occurs in many species and is involved in diverse biological processes. However, the occurrence and role of DNA demethylation in maize remain unknown. Results We analyze loss-of-function mutants of two major genes encoding DNA demethylases. No significant change in DNA methylation has been detected in these mutants. However, we detect increased DNA methylation levels in the mutants around genes and some transposons. The… Show more

“…Bisulfite sequencing of Arabidopsis endosperm and embryo revealed DME-dependent large-scale maternal DNA hypomethylation in the endosperm that correlates with the expression of imprinted genes, showing that DME-directed active DNA demethylation lays down the primary imprinting mark in the endosperm (Gehring et al, 2009;Hsieh et al, 2009;Ibarra et al, 2012). This result is further confirmed in the endosperm of other species such as rice and maize, indicating a widely conserved mechanism in flowering plants Xu et al, 2022).…”

“…In leaves and roots of Brassica napus , methylation differences between homologous gene pairs caused expression bias of homologous genes (Chalhoub et al., 2014). Allele‐specific DNA methylation was found to cause differential expression of alleles during maize endosperm development (Xu et al., 2022). Additionally, DNA methylation (Wang et al., 2015, 2018; Xu et al., 2022; Xue et al., 2022), histone modifications (Guo et al., 2015), non‐coding RNAs function (O'Neill, 2005; Rong et al., 2005), and cis ‐ or trans ‐regulation (Zhang & Borevitz, 2009) are also involved in the ASEs, and further studies are needed to investigate these mechanisms in P. championii .…”

“…This alteration can also take place passively by a reduction or inhibition activity of DNMTs during DNA replication [ 56 , 57 ]. Demethylation is linked to transcription factor binding and proper gene expression [ 58 ]. According to their functions, enzymes that establish, recognize, and remove DNA methylation, such as DNMTs, MBPs, and TET, could be referred to as writers, readers, and erasers, respectively [ 23 , 59 ].…”
