DNA Marker Analysis of ‘Davis’ Soybean and Its Descendants for theRcs3 Gene Conferring Resistance toCercospora sojina

Abstract: Frogeye leaf spot (FLS), caused by Cercospora sojina Hara, is a serious disease of soybean [Glycine max (L.) Merr.]. Complete resistance to all isolates of FLS was reported in the cultivar Davis, and the Rcs3 gene was mapped on Linkage Group J (LGJ), near the simple sequence repeat (SSR) markers Satt244 (64.9 cM) and Satt547 (67.7 cM). The objectives of this study were to track the transmission of Rcs3 from Davis to its descendants and determine the cosegregation of Rcs3 and Davis SSR marker bands. Reaction of… Show more

“…These include 71 isolates from the United States, 15 isolates from Brazil, and 7 isolates from China (Table 1). In total, 93 C. sojina isolates have been analyzed for their reaction on 38 putative soybean differential cultivars (Missaoui et al, 2007). This research was conducted by various individuals over a long time period, and there was a wide range in the number of replications of the various C. sojina isolate–soybean differential evaluations.…”

“…For five more lines, four of the five markers matched the haplotype of Davis, but in each case the marker not matching the Davis haplotype was one of the two markers closest to the Rcs3 gene. The Rcs3 is predicted to be located between AZ573CA150 and Satt‐244 (Missaoui et al, 2007b; Table 3). Remaining lines matched the Rcs3 haplotype of susceptible Blackhawk or had at least three of the five markers matching the haplotype of Blackhawk (data not shown).…”

“…This SSR marker has been successfully used for marker‐assisted selection (MAS) of this allele (Missaoui et al, 2007a). Recently, several single nucleotide polymorphism (SNP) and indel markers that are located within 3 cM of the Rcs3 allele have been identified, confirmed, and used for MAS for Rcs3 (Missaoui et al, 2007b). Marker‐assisted selection with SNP markers is most effective if several SNPs that are evenly spaced and closely linked to the gene of interest are used to define a haplotype of the genomic region flanking the gene (Johnson et al, 2001).…”

“…For PI 594891 and PI 594774, the haplotype windows were defined by the 72.6 kb fine-mapped region (Figs 1 and 2 ). The haplotype window for Davis was defined by the interval based on the primer sequence of Satt244) and AZ573TA150 markers used to map the Rcs3 gene on chr16 [ 30 ]. Therefore, each interval was based on the estimated physical locations of the SSR or SNP markers in the soybean genome map available at www.soybase.org using the Glyma1.01 map version.…”
