DNA methylation in Hepatoblastoma-a literature review

Abstract: Hepatoblastoma (HB) is the most common malignant liver tumor in children. Abnormal activation of the Wnt/βcatenin signaling pathway plays an important role in the formation and development of HB. Genes in HB show a global hypomethylation change, accompanied by hypermethylation of specific tumor suppressor genes (TSGs). This article reviews the hypermethylation changes in several TSGs, such as RASSF1A, SOCS1, APC, HHIP, and P16, and analyzes the pathways and mechanisms of TSGs regulating gene expression. The ro… Show more

The role of these pathways in the development and progression of HB was recently reviewed by Zhang et al [21] . The low rate of mutations in HB together with the alteration of numerous signaling pathways provides evidence of the collaboration of additional mechanisms.…”

“…It is well known that the IGF (insulin-like growth factor) pathway is involved in HB development [21] . The activation of IGF signaling stimulates growth and metastasis in numerous human tumors such as myeloma [53] , HCC [54] , gastric cancer [55] , and lung cancer [56] , among others.…”

“…Hepatoblastoma (HB) is the most common malignant embryonic tumor of the liver in children (most common under 3 years of age), accounting for approximately 79% of all pediatric malignant liver tumors, with an average annual incidence of 1.5 per million population ( 1 3 ). Abdominal masses are major clinical manifestations of HB ( 4 ).…”

“…In the late 1990s, research showed that the existence of mutation of CTNNB1 (β-catenin gene) might lead to β-catenin accumulation, resulting in the development of hepatoblastoma ( 48 ). Some pathways related to hepatoblastoma molecular mechanisms were detected, including the well-studied Wnt/β-catenin and MYC pathways ( 49 ). However, research on the molecular basis in hepatoblastoma was limited.…”
