DNA repair in cancer stem cells as a factor for glioma resistance to radiotherapy

Abstract

The main reason for dismal �rognosis is the high level of tumor recurrence because of resistance to different ways of treatment. currently�� it is believed that glioma develo�ment is connected with the existence of cancer stem cells (cScs��� or tumor-initiating cells. The theory of hierarchal tumor structure is now commonly acce�ted. it accounts for characteristics of these cells�� namely�� the ca�ability of self-renewal and differentiation into astrocytes�� oligodendrocytes�� and neurons. Moreover�� these cells bear multi�le genetic lesions ty�ical of cancer cells. Thus�� the �resence of these cells after surgery and further treatment allows the tumor to recur. The data obtained in recent years confirm the im�ortant role of cScs in the develo�ment of tumor resistance to chemo-and radiothera�y. in this review�� we �resent general information about classification and treatment of gliomas and consider results of research connected with the influence of radiation thera�y. Some authors show that Dna re�air enables cScs to survive even after treatment. To sum u��� it is shown that Dna re�air contributes to the develo�ment of tumor resistance to ionizing radiation. in addition�� our wor� confirms the hy�othesis that inhibition of Dna re�air �rocesses in these cells leads to tumor sensitization to radiothera�y.Key words�� glioma; Dna re�air; radiothera�y; cancer stem cells.Глиомы -опухоли головного мозга�� происходящие из клеток глии и их предшественников. Несмотря на применяемые методы ле�ения�� выживаемост�� пациентов остается крайне низкой. Высокая смертност�� �ол��ных связана с устой�ивост��� этих опухолей к терапии�� в �ол��шинстве слу�аев даже после хирурги�еского удаления опухоли и последу�щего ле�ения сохраняется высокая вероятност�� рецидива за�олевания. В настоящее время развитие глиом связыва�т с так называемыми опухолевыми стволовыми (ОС����� или опухол��-иницииру�щими�� клетками. Широкое распространение полу�ила иерархи�еская модел�� структурной организации опухоли�� при которой вся опухол�� развивается из одной клетки. такие клетки�� о�лада�щие характеристиками стволовых клеток�� за с�ет спосо�ности к самоо�новлени� и дифференцировке�� а также нали�ия генети�еских нарушений о�еспе�ива�т развитие опухоли. таким о�разом�� даже нали�ие не�ол��шого �исла таких клеток после удаления основной массы опухоли может привести к повторному развити� злока�ественной опухоли. Накопленные за последнее время данные свидетел��ству�т о важной роли ОС�� в развитии устой�ивости опухоли к действи� химио-и радиотерапии.
