DNA replication stress and cancer chemotherapy

Abstract: DNA replication is one of the fundamental biological processes in which dysregulation can cause genome instability. This instability is one of the hallmarks of cancer and confers genetic diversity during tumorigenesis. Numerous experimental and clinical studies have indicated that most tumors have experienced and overcome the stresses caused by the perturbation of DNA replication, which is also referred to as DNA replication stress (DRS). When we consider therapeutic approaches for tumors, it is important to e… Show more

“…The transcriptionally active genes, especially those involved in developmental regulation and the cell cycle, interact in the nucleus with the nuclear porin [42]. It is also known that DNA replication disorders cause genomic instability and confer genetic diversity during tumorigenesis [43], explaining how the above pathways may identify effective treatment strategies for LUAD.…”

“…Research in DNA replication is also progressing, and its relationship to cancer development is becoming clearer. Dysregulation of DNA replication can lead to genomic instability, and an important feature of cancer formation is this instability [26] . Cancer may be related to errors in DNA at the replication stage and the combined effects of changes caused by environmental factors and mutations in genetic information [27] .…”
