DNA typing for BoLA class I using sequence-specific primers (PCR-SSP)

Abstract: The analysis of cattle MHC (BoLA) class I gene expression is an essential component of studies on immune responses and susceptibility to disease. International BoLA workshops have generated data and reagents that allow discrimination of class I molecules at the haplotype level, but progress has been limited by difficulties encountered in defining single alleles. Our aim in this study was to develop a DNA-based system for improved identification of expressed class I alleles, utilizing available cDNA sequences d… Show more

“…Four Holstein-Friesian animals (animals 011, 592, 641, and 633) homozygous for the A10 or A18 MHC class I (MHC-I) haplotype were used for the study. Their MHC types were determined by a combination of serological typing (42) and an MHC-I allele-specific PCR (43). The animals were aged 18 to 36 months at the outset of the study and were maintained indoors on rations of hay and concentrate.…”

“…MHC-defined (Ellis et al , 1996, 1998), conventionally reared, 6-month-old Friesian Holstein calves ( Bos taurus ) from The Pirbright Institute (Pirbright) herd were used for these studies. Cannulations were performed essentially as described previously (Hope et al , 2006).…”
