This study aimed to identify novel tyrosinase inhibitory peptides from collagen of donkey by combining in silico screening with in vitro activity verification, and to elucidate inhibition mechanism based on molecular docking and molecular dynamics simulation. Three tripeptides, that is, Asp‐Gly‐Leu (DGL), Gly‐Ala‐Arg (GAR), and Ser‐Asp‐Trp (SDW) were identified and exerted potent tyrosinase inhibitory activities, with IC50 values of 0.47 ± 0.01 mM, 1.13 ± 0.04 mM, and 2.08 ± 0.01 mM, respectively. Each of three identified peptides had hydrophobic amino acids and could stably and closely bind with the active pocket of tyrosinase. Hydrogen bonds played the most important roles in impacting the structure stabilities of the peptide‐tyrosinase complexes. Moreover, His85, His244, His259, and Asn260 were the key residues to drive the interactions between the peptides and tyrosinase. Overall, collagen‐derived peptides DGL, GAR, and SDW from donkey had great potential as tyrosinase inhibitory peptides. Practical Application This study has suggested that three tripeptides DGL, GAR, and SDW derived from collagen of donkey have potent tyrosinase inhibitory activity. These novel collagen‐derived peptides had great potential to be applied as tyrosinase inhibitory peptides to prevent and improve hyperpigmentation disorders and other tyrosinase‐related problems in the food industry. And this work is expected to provide a theoretical basis for the development of novel, safe, and effective tyrosinase inhibitory peptides.

Several studies have shown the association between the ligand-dependent nuclear receptor compression-like protein (LCORL) gene and body size in horses, pigs and donkeys. Based on previous studies, the LCORL gene was hypothesized to be associated with growth traits and hide weight in Dezhou donkeys. In this study, we aimed to reveal the variation of the LCORL gene in the Dezhou donkey and explore whether the gene is associated with hide weight and body size. In this study, genetic polymorphisms in the LCORL gene of the Dezhou donkey were studied using targeted sequencing technology, and single nucleotide polymorphisms (SNPs) of the LCORL gene were analyzed for association with hide weight and body size in Dezhou donkeys. The results showed that there was an SNP locus situated in intron 1 of the LCORL gene. Association analysis revealed that individuals with the GG genotype had significantly higher body height, body length, chest circumference and hide weight than those with the AA genotype (p < 0.05). Therefore, the g.112558859 A > G locus can be used as a potential candidate marker affecting body size and hide weight. This study provides the foundation for breeding high-quality donkeys with high hide yield.

Intramuscular fat (IMF) is an important regulator that determines meat quality, and its content is closely related to flavor, tenderness, and juiciness. Many studies have used quantitative proteomic analysis to identify proteins associated with meat quality traits in livestock, however, the potential candidate proteins that influence IMF in donkey muscle are not fully understood. In this study, we performed quantitative proteomic analysis, with tandem-mass-tagged (TMT) labeling, with samples from the longissimus dorsi (LD) muscle of the donkey. A total of 585,555 spectra were identified from the six muscle samples used in this study. In total, 20,583 peptides were detected, including 15,279 unique peptides, and 2,540 proteins were identified. We analyzed differentially abundant proteins (DAPs) between LD muscles of donkeys with high (H) and low (L) IMF content. We identified 30 DAPs between the H and L IMF content groups, of which 17 were upregulated and 13 downregulated in the H IMF group. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) functional enrichment analysis of these DAPs revealed many GO terms (e.g., bone morphogenetic protein (BMP) receptor binding) and pathways (e.g., Wnt signaling pathway and Hippo signaling pathway) involved in lipid metabolism and adipogenesis. The construction of protein–protein interaction networks identified 16 DAPs involved in these networks. Our data provide a basis for future investigations into candidate proteins involved in IMF deposition and potential new approaches to improve meat quality in the donkey.
